Antibody response to pneumococcal capsular polysaccharide vaccine in Down syndrome patients BJMBR
Costa-Carvalho,B.T.; Martinez,R.M.A.; Dias,A.T.N.; Kubo,C.A.; Barros-Nunes,P.; Leiva,L.; Solé,D.; Carneiro-Sampaio,M.M.S.; Naspitz,C.K.; Sorensen,R.U..
The majority of children with Down syndrome (DS) tend to have frequent bacterial infections including recurrent respiratory infections. Our objective was to evaluate the production of antibodies to pneumococcal polysaccharide antigens after active immunization in DS subjects. IgG antibodies to pneumococcal serotypes (1, 3, 6B, 9V, and 14) were measured before and 6 weeks after immunization with a 23-valent pneumococcal vaccine (Pneumo23®, Pasteur-Merrieux) in 6- to 13-year-old DS children (N = 17) and in aged-matched normal controls (N = 30). An adequate response was defined as a 4-fold increase over baseline or a post-immunization level of specific pneumococcal serotype antibody > or = 1.3 µg/mL. After immunization, all DS children had an increase in...

CD4+ T cells participate in the nephropathy of canine visceral leishmaniasis BJMBR
Costa,F.A.L.; Guerra,J.L.; Silva,S.M.M.S.; Klein,R.P.; Mendonça,I.L.; Goto,H..
Renal involvement in visceral leishmaniasis (VL) is very frequent. The renal lesions of humans and dogs are similar but their pathogenesis has not been clearly elucidated. There is growing evidence that the cellular immune response is involved in the pathogenesis of immunologically mediated glomerulonephritis. Since T cells could participate in the pathogenesis of nephropathy, in the present study we investigated the possible involvement of CD4+ and CD8+ T cells in the nephropathy of canine VL. Six dogs naturally infected with Leishmania (Leishmania) chagasi from the endemic area in the Northeast of Brazil, the town of Teresina in the State of Piauí, were studied. An expressive inflammatory infiltrate of CD4+ T cells both in glomeruli and in interstitium...

Development of an indirect ELISA assay to evaluation of the adaptive immune response of pacu (Piaractus mesopotamicus) Anais da ABC (AABC)
FARIAS,THAIS H.V.; SILVA,KETHERSON R.; MARIGUELA,VIVIANE C.; MONTASSIER,HÉLIO J.; PILARSKI,FABIANA.
ABSTRACT The pacu is one of the most important species for Brazilian fish farming and is considered emerging in the global aquaculture. Despite its importance, no effective tool for evaluation of the adaptive immune response of this species has been developed. Therefore, this study aimed the development and standardization of indirect ELISA for the measurement of pacu antigen-specific antibodies using polyclonal rabbit anti-pacu IgM used as detector antibody. For this purpose was isolated and purificated of pacu IgM using mannose-binding protein affinity chromatography and produced specific polyclonal antibodies against heavy and light chains pacu IgM, that showed a molecular weight of 72 kDa and 26 kDa, respectively. Polyclonal antibodies obtained...

Effect of chronic exposure to ammonia on alterations of proteins and immunoglobulins in sea bass (Dicentrarchus labrax) serum ArchiMer
Coeurdacier, Jean-luc; Dutto, Gilbert.
The relationship between serum protein, immunoglobulin concentrations and protein molecular weight profile (PMWP) alterations of sea bass (131.3 +/- 4.3 g) reared in sea water with sublethal concentrations of ammonia was studied over two periods totalling 116 days. During the exposure period (62 days) the first group (group 1) lived in sea water with 0.204 mg.L-1 unionized ammonia nitrogen equivalent to 12 % of the lethal concentration for 50 % of a population exposed for 96 h to ammonia (96-h LC50), whilst the second group (group 2) lived in sea water with 0.340 mg.L-1 UIA-N equivalent to 20 % of the 96-h LC50 of ammonia. Then, the two groups were left for a recovery period (54 days) in the same water as the control group (group 0). The determination of...

Feeding different levels of vitamin E and selenium has no effect on serum immunoglobulin Y (IgY) production by layers vaccinated against Escherichia coli and avian encephalomyelitis virus Ciência Rural
Kindlein,Giselle; Ribeiro,Andréa Machado Leal; Canal,Cláudio Wageck; Vieira,Maitê de Moraes.
The effects of vitamin E and selenium (Se) supplementation on the immunity of hens vaccinated against a mixture of six swine-pathogenic Escherichia coli (EC) and avian encephalomyelitis virus (AEV) were studied. Antibody production (AbP) was evaluated in ninety 49 to 57-week-old H&N Nick Chick hens fed diets containing 14IU Vitamin E kg-1 (basal diet), 27, 59, 111, or 111IU vitamin E kg-1 + 0.56ppm Se supplementation. At 51 wks of age, half of the hens were vaccinated against EC, and all birds were vaccinated against AEV. At 53-weeks of age, the birds received a second dose of EC vaccine. Blood samples were collected weekly and serum was analyzed by ELISA for anti-EC IgY and was expressed as optical density (OD). Vaccinated hens had higher serum OD...

Le système immunitaire du bar Dicentrarchus labrax. Immunoglobulines, cellules productrices d'immunoglobulines, ontogenèse applications à une pathologie virale néonatale ArchiMer
Breuil, Gilles.
The sea bass, Dicentrarchus labrax, has been subjected to intensive aquaculture on the Mediterranean cost for ten years. Marine fish hatcheries are key factors in the aquaculture field, and face different pathologies, especially one of viral origin (nodavirus) that could challenge their feasibility. Consequently, we have tried to provide some data on larvae and spawning stocks. Firstly, we have studied the immunoglobulins, components of the specific humoral immunity, and their role as antibodies. On adult basses, these immunoglobulins are tetrameric molecules with high molecular weight (850 kD), made of heavy (H) - 80 kD and light (L) - 26 kD chains. In the eggs, simplest forms such as monomeric or dimeric are brought to the fore; they are located under...

Modulation of B lymphocyte function by an aqueous fraction of the ethanol extract of Cissampelos sympodialis Eichl (Menispermaceae) BJMBR
Alexandre-Moreira,M.S.; Piuvezam,M.R.; Pecanha,L.M.T..
Cissampelos sympodialis Eichl species are used in folk medicine for the treatment of asthma, arthritis and rheumatism. In the present study, we investigated the immunomodulatory effect of an aqueous fraction of a 70% (v/v) ethanol extract of C. sympodialis leaves on B lymphocyte function. The hydroalcoholic extract inhibited the in vitro proliferative response of resting B cells induced by LPS (IC50 = 17.2 µg/ml), anti-delta-dextran (IC50 = 13.9 µg/ml) and anti-IgM (IC50 = 24.3 µg/ml) but did not affect the anti-MHC class II antibody-stimulated proliferative response of B cell blasts obtained by stimulation with IL-4 and anti-IgM. Incubation with the hydroalcoholic extract used at 50 µg/ml induced a 700% increase in intracellular cAMP levels. IgM secretion...

Recherche et caractérisation d'immunoglobulines dans les oeufs du loup (Dicentrarchus labrax) ArchiMer
Pepin, Jean-francois; Breuil, Gilles; Rigobert, P; Romestand, Bernard.
The occurrence of maternal immunity transfer in Sea bass (Dicentrarchus labrax) was investigated using ELISA and Western blot technics. In order to determine the possible presence of immunoglobulin (Ig), egg proteins obtained from fertilized eggs were analyzed. After several extraction protocol assays (precipitation, chromatography), monoclonal and polyclonal antibody probes specific to Sea bass Ig allowed to confirm the presence of an immunoglobulin-like protein in eggs with very low level. Biochemical characterization was carried out using SDS-PAGE. Results indicated that egg Ig had a molecular weight of 206 kDa. They were smaller than seric IgM (800 kDa). The heavy (H) and light (L) chains of the egg Ig and of the seric IgM had similar molecular weight,...
